CureGene Begins Human Trials for Oral Metabolic Drug CG-0416

Shanghai-based biotech CureGene has administered the first dose of its experimental drug CG-0416 to a human participant in Australia. The Phase I clinical trial marks the transition of the liver-targeting THR-β agonist from preclinical development to human safety evaluation, targeting obesity and metabolic dysfunction-associated steatohepatitis.

The study is a randomized, double-blind, placebo-controlled investigation assessing the safety, tolerability, and pharmacokinetics of the drug in healthy volunteers. By utilizing Australia’s clinical infrastructure, the company aims to collect data necessary for broader international development. CG-0416 relies on a proprietary prodrug platform designed to release the active compound primarily within the liver, a mechanism intended to reduce systemic side effects.

Preclinical results suggest the drug may offer a distinct advantage in treating metabolic diseases by promoting fat loss while preserving muscle mass—a frequent challenge for patients using current GLP-1 receptor agonists. According to Dr. Gongxin He, founder and CEO of CureGene, the company views this trial as a critical milestone in its expansion into cardiovascular and metabolic therapeutics. The compound also avoids CYP450 oxidative metabolism, which the company claims will minimize drug-drug interactions and improve patient outcomes for long-term daily use.
